Located on the Buffalo Niagara Medical Campus, UB's Proteomics and Bioanalysis Core specializes in proteomics, PK/PD, lipidomics, metabolomics and biopharmaceutical analysis. The Core is a regional resource for UB researchers and regional investigators, providing state-of-the art instrumentation and expertise in proteomics and bioanalysis. Specializing in proteomics, liquid chromatography, mass spectrometry and bioinformatics

Whether your project is large or small, we can help with the identification and quantification of proteins and proteoforms present in a biological sample. Our support can enable insights into the complexities and emergent properties of biological systems for drug discovery and development activities. After an initial discussion of your requirements, we will confirm how we can help. If you'd like to proceed, we'll assign an expert team and project manager. 

The facility has several main research foci: 

• Ultra-sensitive quantification of small-molecule analyses, such as drugs, metabolites and biomolecules 

• Comprehensive protein expression profiling and differential proteome analysis 

• Ultra-sensitive quantification of proteins/peptides

Protein identification

  • Digestion with trypsin or alternative enzymes 
  • Sample preparation with RPLC and retention time determination in chromatogram 
  • Peptide fragmentation (HCD) and m/z determination in MS/MS spectrum

Protein characterization

  • Post-translational modifications (PTMs): phosphorylation, deamidation, methylation, nitration, acylation 
  • Other: cleavage site determination, amino acid substitution, disulfide crosslinking, chemical modifications

Technologies

Micro-scaffold Assisted Spatial Proteomics (MASP)

Ion-current-based MS1 global proteomic quantification Ion (IonStar) - an MS1-based quantitative method devised for large-cohort proteomics analysis, which prominently alleviates issues associated with quantitative precision, missing data, and false-positive discovery of protein changes.

State-of-the art instrumentation

The Core consists of two instrument rooms, totaling 600 sq ft, which house a number of state-of-the-art liquid chromatography (LC) systems, mass spectrometer (MS), and sample processing instruments: 

  • Thermo Orbitrap Astral mass spectrometer
  • Thermo TSQ Altis triple quadrupole mass spectrometer
  • Thermo TSQ Quantiva triple quadrupole mass spectrometer
  • Thermo Dionex Ultimate 3000 nano/micro HPLC systems with WPS-3000 autosamplers
  • Waters Bioseparation semi-preparation HPLC system
  • Several additional nano/micro LC systems (i.e. Eksigent nano-2D, Agient 1200-series micro HPLC, Shimadzu AC20 HPLC)
