Dr. Huang`s research focuses on the regeneration of adult oligodendrocyte precursor cells (OPCs). Following CNS demyelination,OPCs are able to perform the regenerative task of replacing myelin in the brain. To understand how genes and signaling pathways regulate CNS remyelination, he used a model of experimental demyelination in rats followed by laser capture microdissection of lesions and microarray/bioinformatic analysis to generate a comprehensive gene expression profile of CNS remyelination. He found that a novel gene, retinoid X receptor-gamma (RXRgamma) is highly upregulated in the lesioned CNS, and is required for adult OPC differentiation to regenerate myelin (Huang et al., Nature Neurosci, 2011). Moreover, he found that RXR agonists were able to enhance CNS remyelination in vivo. Since drugs that target RXR signaling (rexinoids) are available, and are currently being tested for treatment of certain cancers (by differentiation therapy) and metabolic diseases, it is possible that rexinoids could also be used as regenerative therapeutics to stimulate remyelination in demyelinating diseases such as multiple sclerosis (Huang and Franklin, Neurochem Int, 2011; Huang et al., Expert Rev Neurother, 2011).
